SOP Of SWAB TEST

 

OBJECTIVE:To define the procedure for detection of bioburden by swab test

 

SCOPE: This SOP is applicable for detection of bioburden by swab test in Microbiology Laboratory.

 

RESPONSIBILITY:Microbiologist: to follow the process given as per SOP.

Q.C Head: to ensure the proper execution of the SOP.

Q.A: to ensure that the SOP is being properly implemented.

ABBREVIATION:

SCDM- Soyabean casein digest agar

SCA- Saboraud’s chloramphenicol agar

                                                                                                                                   

PROCEDURE:

 

  1. Sterilize the Johnson & Johnson buds (ready made swabs) by autoclaving at 121º C, 15 lbs for 20 minutes.
  2. Put the above swab in a test tube containing about 1 to 2 ml saline (ensure that the swab gets moistened).
  3. Streak cotton swabs on about 400 cm2 area surface of the laminar air flow Cabinet/other surfaces of which bioburden is to be tested.
  4. Deep the cotton swabs in 10 ml. of sterile soyabean casein digest medium or nutrient broth.
  5. Take 1 ml. of above solution in sterile Petri dish and pour previously sterilized & cooled 15-18 ml SCDA medium for total bacterial count.
  6. Take 1 ml. of above solution in sterile Petri dish and pour previously sterilized & cooled 15-18 ml SCA medium for total fungal count.
  7. Allow the plates to solidify at room temperature for half an hour.
  8. Incubate the SCDA plates at 30-350 C for 5 days and SDA plates at 20-250 C for 5 days.
  9. Count the No. of colonies multiply with dilution factor (10) and express the results as Colony Forming Unit (cfu)/400 cm2.

 

LIMITS:

Total bacterial count: NMT 100 cfu / 400 cm2

Total fungal count: NMT 10 cfu/400 cm2

Annexure I: Swab testing report

 

Media      1    :  Soyabean Casein Digest agar for Total Bacterial Count (TBC) and

2    :  Sabouraud Dextrose agar with antibiotic for Total Fungal Count (TFC).

  Incubation    : 120 hrs. at 30 to 35°C for Total Bacterial Count (TBC)  and  120 hrs at 20 to 25°C for Total Fungal Count (TFC).
